Transport Kinetics of Succinate in Renal Basolateral membrane Vesicles of Cadmium-Intoxicated Rabbits


Abstract
Changes in transport kinetics of succinate were studied in renal basolateral membrane vesicles(BLMV) isolated from cadmium-intoxicated rabbits. Cadmium intoxication was induced by subcutaneous injections of CdCL2 at a dose of
3mg/kg.

over 3 weeks. Basolateral membrane vesicles were prepared from renal cortex by Percoll gradient centrifugation. Cadmium intoxication resulted in marked attenuation of Na+-dependent transport of succinate in basolateral me membrane
vesicles,
and this was due to reduction in Vmax and to increase in Km.
These results indicate that cadmium intoxication impairs the Na+-succinate cotransport system in the basolateral membrane of renal proximal tubular cells.
